Effect Of Psoralen Extract On Differentiation Of Mouse Osteoblast Cell Line Mc3T3-E1 Cells Cultured In Vitro

Abstract
Objective: To study the effect of psoralen extract on the differentiation of neonatal mouse calvarial MC3T3-E1 cells cultured in vitro. Methods: The mouse osteoblast cell line MC3T3-E1 was used as a cell model for drug screening. The cultured MC3T3-E1 mouse osteoblasts were divided into 4 groups: blank group, psoralen extract (containing crude drug) 0.02, 0.2 , 2g¡¤L-1 different dosage groups. The contents of cell ALP and type I collagen were detected, and real-time PCR was used to determine the effect of psoralen extract on the expression of ALP, type I collagen and osteocalcin mRNA in MC3T3-E1 cells. Results: Psoralen extract crude drug dose 0.2g¡¤L-1 can increase the activity of ALP in MC3T3-E1 cells and promote the secretion of type ¢ñ collagen. At the same time, it significantly promoted the expression of ALP in MC3T3-E1 cells from 7 to 14 days, and the mRNA expression of type I collagen and osteocalcin from 14 to 21 days. Conclusion: The traditional Chinese medicine Psoralen extract can effectively promote the differentiation of osteoblasts and provide a reference for studying the mechanism of action of the traditional Chinese medicine Psoralen.
