Abstract
Study the effect of microgravity on the promoter activity of COL1A1 (type I collagen ¦Á1 chain gene), and explore the mechanism of microgravity’s effect on the expression of osteoblast-related genes. The 3.6kb COL1A1 promoter was double-digested to obtain promoters of different lengths. The sub-fragment was connected to the reporter gene EGFP (enhanced green fluorescent protein), transfected into ROS17/2.8 cells, and screened with G418 to obtain a ROS17/2.8 cell line stably transfected with the COL1A1-EGFP gene. A rotator was used to simulate the microgravity effect. , under in vitro culture conditions, the expression of reporter genes of each cell line was observed. The results showed that after the cells were cultured under simulated microgravity for 24 and 48 hours, the expression of the reporter gene EGFP and type I collagen increased, indicating that the activity of the COL1A1 promoter was enhanced. Description Under short-term simulated microgravity conditions, osteoblasts can compensatoryly increase the expression of type I collagen by enhancing COL1A1 promoter activity.
