Abstract
Objective To observe the effect of achyranthes bidentata combined with ultrasound treatment on insulin-like growth factor-1 (IGF-1) and type II collagen in the knee joint cartilage tissue of rabbits with knee osteoarthritis (OA) model, and to preliminarily explore its mechanism of preventing and treating OA. Methods New Zealand white rabbits were divided into normal group, model group, Achyranthes bidentata group, ultrasound group and observation group using random number table method. According to the Hulth modeling method, experimental rabbits in the model group, Achyranthes bidentata group, ultrasound group and observation group were made into knee OA animal models. One week after the modeling, the Achyranthes bidentata group and the ultrasound group were given Achyranthes bidentata and ultrasound treatment respectively, while the observation group was given combined treatment of Achyranthes bidentata and ultrasound. The materials were collected after 4 weeks of treatment, and immunohistochemistry was used to detect the contents of IGF-1 and type II collagen in the cartilage tissue of each group of experimental rabbits, and the differences between the groups were compared through the image analysis system. Results The expression of IGF-1 in the cartilage tissue of the Achyranthes bidentata group, ultrasound group and observation group was significantly enhanced compared with the model group (P<0.05); the difference in IGF-1 expression between the ultrasound group and Achyranthes bidentata group was statistically significant (P>0.05). The expression of IGF-1 in the observation group was significantly higher than that in the Achyranthes bidentata group and ultrasound group (P<0.05). The expression of type II collagen in the cartilage tissue of the Achyranthes bidentata group, ultrasound group and observation group was significantly enhanced compared with the model group (P<0.05); the difference in the expression of type II collagen between the ultrasound group and the Achyranthes bidentata group was statistically significant (P>0.05), observation The expression of type II collagen in the group was significantly enhanced compared with the Achyranthes bidentata group and the ultrasound group (P<0.05). Conclusion The traditional Chinese medicine Achyranthes bidentata combined with ultrasound treatment can significantly increase the expression of IGF-1 and type II collagen in the cartilage tissue of OA experimental rabbits.
