Abstract
¡¾Abstract Objective? ? To explore the effects of activin A (ACT-A) and follistatin (FS) on the synthesis of type I collagen (ColI) by rat renal interstitial fibroblasts cultured in vitro. square? Law? ? The renal medulla of SD rats was removed under sterile conditions and cultured by collagenase digestion method. After being identified as renal interstitial fibroblasts, they were divided into 3 groups: different ACT-A concentration groups (group A), no? The same FS concentration group (F group), 30ng/ml ACT-A + different concentration FS group (A+F group), immunohistochemistry and RT-PCR were used to detect ColI protein and ColImR-? Expression of NA. result? ? Rat renal interstitial fibroblasts were successfully cultured. Within the concentration range of this study, ACT-A promoted the expression of ColI protein (P??<0.01) and ColImRNA (P<0.01) in rat renal interstitial fibroblasts. FS can inhibit the above-mentioned effects of 30ng/ml ACT-A (P<0.01), while simple FS can inhibit rat renal interstitial fibroblast ColI? The expression of protein and mRNA had no effect (P>0.05). in conclusion? ? ACT-A may play an important role in the occurrence and development of renal interstitial damage. Is it possible to supplement exogenous FS as a treatment? Provide new ideas for treating renal interstitial fibrosis.
