Abstract
Objective To study the effect of black coffee extract on collagen synthesis and aging-related genes in human skin fibroblasts (HSF). Methods: HSF cells were cultured in vitro, and the safe dose of black coffee extract was screened by CCK-8 method; the experiment was divided into sample group and blank control group, and ELISA technology was used to detect the effect of black coffee extract on type ¢ñ collagen (collagen ¢ñ, COI ¢ñ) in the cells. ) and matrix metalloproteinase-1 (MMP-1) content, RT-PCR technology was used to detect the expression levels of COI ¢ñ and MMP-1 mRNA in cells after black coffee extract. Results Compared with the blank control group, black coffee extract could inhibit the synthesis of MMP-1 in HFF-1 cells (P<0.05), promote the synthesis of COI ¢ñ (P<0.05), and increase the expression level of COI ¢ñ mRNA (P<0.01 ), while the expression level of MMP-1 mRNA decreased (P<0.01). Conclusion Black coffee extract can improve and regulate collagen metabolism by promoting CoI I and inhibiting MMP-1 expression, and can delay skin aging.
