Abstract
Objective To observe the effects of dehydrocordine (DHC), the main active ingredient of the traditional Chinese medicine Corydalis Corydalis, on the proliferation and collagen secretion of cardiac fibroblasts (CFs) cultured with high concentration of glucose, so as to provide experimental basis for the clinical application of Corydalis Corydalis. Methods The CFs model cultured in high glucose in vitro was used for experiments. The CFs of suckling mice within 24 hours after birth were isolated by combined digestion with collagenase and trypsin. The cells from the 2nd to 4th generations were cultured in high-concentration glucose (25 mmol/L), and 100, 50, and 25 mg/L DHC were added respectively for intervention. 24, 48 Cell morphology was observed under the microscope after h, cell proliferation was detected by MTT method, cell cycle was detected by flow cytometry, and the content of type I and type III collagen was determined by ELISA. Results CFs began to grow adherently after 3 hours of transplantation. Cell proliferation increased significantly after 24 and 48 hours of culture in high-concentration glucose culture medium (P£¼0.05, P£¼0.01). After 48 hours, the percentage of cells in the S+G2+M phase increased significantly (P£¼0.01 ), the amount of type ¢ñ and type ¢ó collagen secreted by cells increased significantly (P£¼0.01); DHC intervention can significantly inhibit the proliferation of CFs (P£¼0.01), reduce the percentage of cells in the S+G2+M phase (P£¼0.01), and reduce the number of types ¢ñ and ¢ó. Collagen secretion (P£¼0.05, P£¼0.01). Conclusion DHC can inhibit the proliferation of CFs cultured in high glucose and reduce collagen secretion. DHC has potential anti-myocardial fibrosis effect.
