Abstract
Objective: To explore the effect of Eucommia polysaccharide in treating liver fibrosis (HF) and explore its related mechanism of action. Methods: 60 healthy SD rats were randomly divided into 2 groups, the normal group (10 rats) and the liver fibrosis model group (50 rats). In the modeling group, 40% carbon tetrachloride (CCl4) was intraperitoneally injected to prepare the HF animal model. After successful modeling, they were randomly divided into 5 groups, namely the model group, Eucommia polysaccharide high, medium and low dose groups and Colchicum There are 10 animals in each group in the alkaline group. Samples were collected from the colchicine group (1.0¡Á10-4g¡¤kg-1) and Eucommia polysaccharide high-, medium-, and low-dose groups (0.14, 0.07, 0.035 g¡¤kg-1) after continuous intragastric administration for 8 weeks, respectively. The body weight of rats and liver coefficient were calculated; enzyme-linked immunosorbent assay (ELISA) was used to detect serum interleukin-6 (IL-6), high-mobility group box 1 (HMGB1), lipid polysaccharide (LPS), liver tissue matrix metalloproteinase-1 (MMP-1) and tissue inhibitor of metalloproteinase-1 (TIMP-1) levels; reverse transcription polymerase chain reaction (RT-PCR) technology analysis of rats in each group Expression of type I and type III collagen, MMP-1, TIMP-1 and transforming growth factor-¦Â1 (TGF-¦Â1) mRNA in liver tissue. Results: Compared with the normal group, CCl4 could significantly reduce the body weight of the experimental rats (P<0.01) and increase the liver coefficient (P<0.01); Eucommia polysaccharide could significantly increase the body weight of the HF model (P<0.01) and significantly reduce the HF Model liver coefficient (P<0.01); ELISA test showed that Eucommia polysaccharide can significantly reduce serum IL-6, HMGB1 and LPS contents in rats with liver fibrosis (P<0.01); RT-PCR test results showed that Eucommia polysaccharide and colchicum Alkali can significantly reduce the contents of type ¢ñ and type ¢ó collagen, TIMP-1 and TGF-¦Â1 mRNA in HF liver (P<0.05), and significantly increase the content of MMP-1 (P<0.05, P<0.01), with a dose-effect relationship. . Conclusion: Eucommia polysaccharide has significant anti-hepatic fibrosis effect, and its mechanism may be related to inhibiting and reducing the expression of collagen type I and type III, TIMP-1 and TGF-¦Â1 mRNA in HF liver.
