Abstract
Objective: To observe the anti-pulmonary fibrosis mechanism of Fuzheng Huayu Recipe on MMP-2 activity and type IV collagen expression in lung tissue. Methods: A one-time intratracheal injection of bleomycin was used to induce pulmonary fibrosis model, which was divided into model control group, Fuzheng Huayu Recipe group, and methylprednisolone group, with 14 model rats in each group. Another normal group of 10 rats was established, and the same amount of normal saline was injected into the trachea at one time. On the day after modeling, the Fuzheng Huayu Recipe group was given 4.6 g¡¤kg-1 rat weight of Fuzheng Huayu Recipe by gavage; the methylprednisolone group was given 15 mg¡¤kg-1 rat weight of methylprednisolone intraperitoneally. The normal group and The model group was fed an equal amount of normal saline once a day for a total of 4 weeks. Masson staining was used to observe collagen deposition in lung tissue, lung weight was measured and lung coefficient was calculated, hydroxyproline content in lung tissue was determined by Jamall’s method, activity of MMP-2 and MMP-9 in lung tissue was detected by gelatinase diagram, and lung tissue was detected by Western blot. Protein expression of MMP-2, MMP-9 and type IV collagen. Results: Compared with the normal group, the lung coefficient of the model group increased significantly, the collagen deposition in the lung tissue was obvious, the Hyp content, type IV collagen, MMP-2 and MMP-9 protein expression increased, and the MMP-2 activity level increased, especially the active type. MMP-2 activity increased significantly, while MMP-9 activity decreased slightly. Compared with the model group, Fuzheng Huayu Recipe and methylprednisolone reduced collagen deposition and type IV collagen expression in lung tissue of the model group to varying degrees, reduced lung coefficient and Hyp content in lung tissue, and reduced MMP-protein and 2 activity levels, especially Active MMP-2 levels. Among them, Fuzheng Huayu Recipe has the most obvious inhibitory effect on lung coefficient, type IV collagen and MMP-2 protein and activity. The two drugs had no significant effect on MMP-9 protein expression and activity. Conclusion: Fuzheng Huayu Recipe has a good anti-pulmonary fibrosis effect in rats, and part of its mechanism of action is: inhibiting MMP-2 protein and its activity in fibrotic lung tissue to reduce tissue inflammatory destruction; inhibiting excessive type IV collagen expression to reduce the pathological proliferation and deposition of collagen.
