Abstract
Abstract: Objective To explore the effect of IL-17 on the proliferation, transformation and collagen synthesis of lung fibroblasts in the formation of bleomycin-induced pulmonary fibrosis. Methods: Intratracheal infusion of 5 mg/kg bleomycin was used to induce pulmonary fibrosis. Reverse transcription-polymerase chain reaction (rt-PCR) was used to detect the expression changes of il-17/il-17rmRNA in the formation of pulmonary fibrosis. Lung fibroblasts expressing IL-17RMRNA at the optimal time were selected for primary culture and identification. The proliferation of lung fibroblasts induced by different concentrations of IL-17 was detected using tetrazolium blue colorimetric assay. RT-PCR and western blotting were used to detect the expression of myofibroblast marker ¦Á-SMA and types I and III collagen. Results (1) il-17/il-17rmRNA was significantly increased in the formation of pulmonary fibrosis, with the highest expression on the 14th day. (2) Different concentrations of il-17 can promote the proliferation of lung fibroblasts, and the optimal concentration is 50ng/ml. (3) IL-17 promotes the increase of ¦Á-sma expression in lung fibroblasts. (4) IL-17 promotes the synthesis of type I and III collagen in lung fibroblasts. Conclusion IL-17 can promote the proliferation, transformation and collagen synthesis of lung fibroblasts in the formation of pulmonary fibrosis in mice, and may play an important role in the formation of pulmonary fibrosis.
