Effects Of Isopsoralen On Collagen In Mouse Embryonic Preosteoblasts

Abstract
Objective To explore the effects of different concentrations of isopsoralen on the proliferation of mouse embryonic preosteoblasts (MC3T3-E1), the expression levels of type I collagen and AP-1 subunit c-Jun and its relationship with primary bone quality Porosis relationship. Methods Cells were divided into normal control group and different concentrations of isopsoralen groups. MTT method was used to detect the proliferation of osteoblasts (MC3T3-E1), Sirius red staining method was used to detect the collagen content of the cell layer, Western-Blot was used to detect c-Jun and RT-PCR was used to detect the expression of type I collagen mRNA. Results Stimulation with different concentrations of isopsoralen can significantly promote the proliferation of MC3T3-E1, increase the collagen content of the cell layer, gradually enhance the expression of c-Jun protein and increase the expression of type ¢ñ collagen mRNA, and the differences are statistically significant (P<0.05 ). Conclusion Isopsoralen may play its role in preventing and treating osteoporosis by promoting the expression of nuclear transcription factor c-Jun, thereby promoting collagen synthesis.
