Abstract

Objective To explore the possible connection between macrophage migration inhibitory factor (mif) and collagen synthesis by vascular smooth muscle cells (vsmc). Methods: The rat aortic media smooth muscle was cultured as primary cells. The 4th passage cells were divided into two groups. Mif at 25, 50, 75 and 100ng/ml was used to stimulate the growth of vsmc to form the experimental group. Mif was not used to stimulate the growth of vsmc. For the experimental group, RT-PCR and PCR methods were used to determine the vsmc collagen ¢¡ and ¢£ mRNA content. Results Compared with the normal control group, except for 25ng/ml, the other three different concentrations of mif stimulated SMC and the relative content of collagen I mRNA was significantly increased (p<0.05). When 100ng/ml mif stimulated SMC, the relative content of collagen III mRNA was significantly increased compared with the control group (p<0.05), while the relative content of collagen III mRNA at other concentrations had no significant difference compared with the control group. Conclusion There is a relationship between mif and vsmc collagen synthesis. Mif stimulation can promote the expression of collagen ¢¡ and ¢£ mRNA in vsmc cultured in vitro.
