Effects Of Mir-144 Overexpression Lentivirus On The Pathological Progression Of Synovial Tissue In Arthritis Model Mice

Abstract
Objective To explore the effect of miR-144 overexpression lentivirus on the pathological process of synovial tissue in arthritis model mice and its possible mechanism. Methods: Type II collagen emulsion was injected subcutaneously into the tail base of BALB/c mice to construct a mouse model of collagen-induced arthritis (CIA), and then a lentivirus with high expression of miRNA-144 was injected into the tail vein. Then, immunohistochemistry was performed on the synovial tissue of the model mice to detect the expression and distribution of Cadherin-11 and TNF-¦Á, and HE staining was used to detect the pathological changes in the synovial tissue. Results The synovial tissue of the model mice showed similar characteristics to the pathological process of rheumatoid arthritis. The inflammatory response of the synovial tissue of the model mice treated with lentivirus with high expression of miRNA-144 was compared with that of the saline and empty lentivirus treated groups. The expression of TNF-¦Á was significantly reduced compared with the normal saline and empty vector lentivirus treatment groups (P<0.05). Further detection of the expression of Cadherin-11 was also significantly reduced (P<0.05). Conclusion Increased miRNA-144 in the body can attenuate or inhibit the inflammatory response of synovial tissue in arthritis model mice and downregulate the expression of TNF-¦Á, and this effect may be due to the inhibition of Cadherin-11 in synovial cells by miRNA-144. effect.
