Abstract
Objective To study the effect of lipopolysaccharides (LPS) on the proliferation of rat glomerular mesangial cells and the secretion of extracellular matrix as well as the protective effect of probucol (PRB). Methods After synchronization, the cells were divided into: normal control group; LPS group (LPS 10 ¦Ìg?mL-1); PRB+LPS treatment group (PRB concentrations were 10, 20, 50 ¦Ìmol?L-1 respectively), and continued to be cultured for 24. 48,72 h. The proliferation of mesangial cells was detected by MTT method, the expression difference of type IV collagen (college-IV, COL-IV) and connective tissue growth factor (CTGF) mRNA was detected by RT-PCR method, and the expression difference of type IV collagen (COL-IV) and connective tissue growth factor (CTGF) mRNA was detected by Western blot method. Differences in expression of COL-IV and CTGF proteins. Results After 24, 48, and 72 hours of treatment, cell proliferation in the LPS group was significantly increased compared with the control group (P<0.05). Compared with the LPS group, cell proliferation in the PRB+LPS group was significantly inhibited (P<0.05), in a time-dose dependence. Effect (r=0.932, P<0.05). After 48 hours of culture, the secretion of COL-IV and CTGF in the LPS group was significantly increased compared with the control group (P<0.05); after pre-treatment with different doses of PRB, the levels of COL-IV and CTGF mRNA and protein were down-regulated compared with the LPS group ( P<0.05). Conclusion LPS can promote mesangial cell proliferation, induce the release of cytokines, promote the synthesis of extracellular matrix, and participate in the occurrence of lipotoxic renal damage. PRB may exert a renal protective effect by downregulating the expression of these vascular growth factors.
