Abstract
Objective To explore the therapeutic value and possible mechanism of Tongxinluo on diabetic cardiomyopathy by observing the effect of Tongxinluo on myocardial proliferating cell nuclear antigen (PCNA) in streptozotocin-induced diabetic rats. Methods Among 60 SD rats, 17 were randomly selected as the control group; 40 diabetic rats that were successfully modeled were randomly divided into 4 groups: model 8-week, 12-week group and Tongxinluo treatment 8-week, 12-week group. Each group 10 rats, two groups of Tongxinluo were given Tongxinluo after successful self-modeling. The experiment monitored blood sugar every 4 weeks. At the two experimental end points of 8 weeks and 12 weeks, rat ventricular muscle tissue was taken for immunohistochemistry detection. Proliferating cell nuclear antigen percentage (PCNA%) and myocardial collagen percentage. Results The blood glucose levels of rats in the model group and Tongxinluo treatment group were significantly higher than those in the control group (P0.05); myocardial PCNA% in the 8-week and 12-week model groups increased compared with the control group (P<0.01); Tongxinluo treatment group Myocardial PCNA% in the two treatment groups was significantly reduced compared with the model group at the corresponding time point (P<0.01). At the 8th and 12th weeks of the experiment, the expression of myocardial collagen in the model group was significantly higher than that in the control group (P<0.01). The expression of myocardial collagen in the Tongxinluo treatment group at 8 weeks and 12 weeks was significantly lower than that in the model group at the corresponding time points (P<0.05). Conclusion Tongxinluo can reduce the expression of PCNA and fibrosis in myocardial tissue of diabetic rats.
