Abstract
Objective: To observe the effects of total saponins of Thladiantha dubia root (TSTR) on the osteoclast (OC) positive feedback factor leukocytes in blood and knee joint synovium in rats with bovine type II collagen-induced arthritis (CIA). Changes in levels of interleukin-15 (IL-15) and IL-34 to explore the mechanism of action of TSTR in treating CIA in rats. Methods: There were 80 male Wistar rats, 10 of which were randomly selected as the normal group. The remaining rats were injected with an emulsion of bovine type II collagen mixed with complete Freund’s adjuvant into the skin of the tail root and back at multiple points. The same method was performed 7 days later. 14 days after the first injection, the rat arthritis index (AI value) ¡Ý4 was considered as successful modeling. The model rats were randomly divided into model group, TSTR low, medium and high dose groups ( 20, 40, 80 mg¡¤kg-1) and Tripterygium wilfordii polyglycoside (TG)-positive drug group (12 mg¡¤kg-1). Each drug group was given the corresponding drug by gavage for 35 days, once a day. The swelling degree of rat paws was observed and the AI value was evaluated; hematoxylin-eosin staining (HE) was used to observe the pathological changes of the synovium; enzyme-linked immunosorbent assay (ELISA) was used to detect IL-15 and IL-34 in rat serum. content; reverse transcription-polymerase chain reaction (RT-PCR) was used to detect IL-15 and IL-34 mRNA expression levels in rat synovium; Western blot was used to detect IL-15 and IL-34 in synovium. protein expression levels. Results: Compared with the normal group, the AI value of rats in the model group was significantly higher (P<0.01), the number of inflammatory cells in synovial cells increased, and the contents of IL-15, IL-34, Gene and protein expression were significantly increased (P<0.01); compared with the model group, the AI values of each dose group of TSTR and the TG group were significantly reduced (P<0.01); and the number of inflammatory cells in the synovium of the TSTR low-dose group and the TG group and the number of blood vessels was significantly reduced; the expression of IL-15 and IL-34 in the serum of rats in the TSTR low-dose group and the TG group, and the expression of IL-15 and IL-34 protein in the synovium, as well as the expression of IL-15 and IL-34 mRNA. Significantly reduced (P<0.05, P<0.01). Conclusion: The therapeutic effect of TSTR on CIA rats may be related to the inhibition of the expression of IL-15 and IL-34 in blood and synovial tissue.
