Abstract
[Abstract] Objective: The traditional Chinese medicine puffball acts on fibroblasts of rat skin cultured in vitro, observes the proliferation and collagen synthesis abilities of rat fibroblasts, and explores the mechanism of the effective components of puffball in promoting the healing of skin lesions at the cellular level. . Methods: In vitro cell culture technology was used to culture fibroblasts from the skin of neonatal rats. The experiment was divided into control group (no administration group) and puffball effective component 37.5, 75.0, 150.0 and 300.0mg/L groups; In the experimental group, different concentrations of drugs were added 24 hours after subculture, and cells were collected after continuing to culture for 24, 48, and 72 hours. MTT colorimetric method was used to determine cell proliferation, and biotin double-antibody sandwich enzyme-linked immunosorbent assay was used to determine cellular collagen content. Results: Compared with the control group, the growth rate of fibroblasts and the collagen synthesis capacity of fibroblasts in the 37.5, 75.0, 150.0 and 300.0mg/L experimental groups were significantly increased (P<0.01). In terms of drug action time, compared with the control group, the cell proliferation rate and collagen synthesis ability were significantly enhanced at 24, 48 and 72 hours, with the strongest effect at 72 hours, and the difference was statistically significant (P<0.01). Conclusion: Under the action of different doses of puffball effective components, rat fibroblasts are activated and proliferated, and their ability to synthesize collagen is enhanced.
