Abstract
Objective To explore the effects of receptor tyrosine kinase inhibitors on chondrocytes at the cellular and molecular level and their possible mechanisms through experimental studies on cultured chondrocytes in vitro. Methods Primary and subcultured chondrocytes were cultured. Chondrocytes suitable for subculture were selected through observation of cultured cell morphology and toluidine blue staining. RT-PCR and ICC were performed 1, 2, 3, and 4 days after trihydroxyisoflavone intervention. and toluidine blue staining observation method to compare the changes in drug intervention on the transcription and translation levels of type I and type II collagen and the expression of proteoglycans in chondrocytes cultured in vitro. Results 20 ¦Ìg/mL and 25 ¦Ìg/mL trihydroxyisoflavone intervention could inhibit the expression of type ¢ñ collagen and increase the expression of type ¢ò collagen and proteoglycan. Conclusion Trihydroxyisoflavones can inhibit the reverse differentiation of chondrocytes induced by lipopolysaccharide stimulation, and have a certain protective effect on the normal phenotype of articular chondrocytes cultured in vitro. Its protective effect has a certain correlation with the concentration of trihydroxyisoflavones.
