Abstract
Objective To observe the direct effects of urotensin ¢¢ (u¢¢) on the proliferation of cardiac fibroblasts (cfs) and the expression of type ¢¡ collagen (col-¢¡) gene in neonatal mice in vitro, so as to explore the role of urotensin ¢¢ in myocardial remodeling in heart failure. role. Methods: CFS from SD suckling mice were isolated and purified by trypsin digestion and differential adhesion method. Cell proliferation was measured by tetrazolium salt (MTT) colorimetric method, and col-¢¡mRNA expression level was measured by RT-PCR. Results: u¢¢ has a concentration-dependent effect on cfs proliferation. As the concentration of u¢¢ increases, the mtt light absorption value first increases and then decreases. Among them, the effects of u¢¢ on 1¡Á10-8mol/l and 1¡Á10-9mol/l Significantly (p<0.05); UII at concentrations of 1¡Á10-7mol/l, 1¡Á10-8mol/l and 1¡Á10-9mol/l can promote col-¢¡mRNA expression in cfs (p<0.01). Conclusion u¢¢ can directly promote the proliferation of cfs and the expression of col-¢¡ mRNA, and the magnitude of the effect is related to the concentration of u¢¢, suggesting that u¢¢ may play an important role in the pathophysiological process of myocardial remodeling in heart failure.
