Abstract
Objective To observe the effect of Longzuan Tongbi Recipe, a Zhuang medicine, on the serum and synovial Fas/FasL systems of rats with collagen-induced arthritis (CIA). Methods 10 of 60 male Wistar rats were randomly selected as the normal control group, and the remaining 50 were induced with a mixture of bovine II collagen and Freund’s incomplete adjuvant to create a CIA model. After modeling, they were divided into model group and methotrexate group. group (MTX group), high-, medium-, and low-dose groups of Zhuan Tongbi prescription (referred to as high-, medium-, and low-dose Zhuang drug groups), with 10 animals in each group. The model group was gavaged with normal saline, and the MTX group was gavaged with MTX solution at 0.27 mg/100 g of rat body weight, once a week for 4 weeks; the high-, medium-, and low-dose aphrodisiac groups were given the aphrodisiac drug Longzuan Tongbi respectively. The decoction (4.32, 2.16, 1.08 g/mL) was administered twice a day for 30 days. HE staining was used to observe the morphological changes of synovial membrane; ELISA method was used to quantitatively detect the expression of Fas/FasL in rat serum and synovial tissue homogenate after intervention. Results In the normal group, there were normal synovial cells. In the model group, synovial cells proliferated significantly, the lower fat cells were reduced or deformed, and fibroblasts increased, accompanied by lymphocyte and monocyte infiltration; in the MTX group and those with low, medium, and The high-dose group was significantly improved compared with the model group. Compared with the normal group, the expression of Fas in the serum and synovial tissue of the model group increased, and the expression of serum FasL decreased (P<0.05, P<0.01); The expression of Fas in the tissue decreased, and the expression of FasL in the synovial tissue of the MTX group, the low-, medium-, and high-dose groups of the aphrodisiac and the synovial tissue of the MTX group, the medium-dose and high-dose groups of the aphrodisiac increased (P<0.05, P<0.01); compared with the MTX group , the expression of Fas in the serum and synovial tissue of the low- and medium-dose groups of the strong-enhancing drug increased, the expression of Fas in the serum and synovial tissue of the high-dose group of the strong-enhancing drug decreased, and the expression of Fas in the serum and synovial tissue of the low- and medium-dose groups of the strong and strong drug increased. The expression of FasL decreased, and the expression of FasL in the serum and synovial tissue of the high-dose aphrodisiac group increased (P<0.05, P<0.01). Conclusion Regulating the apoptosis mechanism mediated by the Fas/FasL system and slowing down the pathological reactions of rheumatoid arthritis may be one of the mechanisms of the Zhuangmao Longzuantongbi prescription in controlling and treating rheumatoid arthritis.
