Abstract
Abstract: Objective To observe the immunomodulatory effect of chinonol on bovine type II collagen-specific lymphocytes in vivo and in vitro, and to explore the immunosuppressive mechanism of chinonol on antigen-specific lymphocytes. Methods Wistar rats were randomly divided into control group, model group, chinonol group and prednisolone group. Except for the control group, the collagen-induced arthritis rat model was replicated and administration started on the day of modeling. After 30 days of modeling, the spleen and draining lymph nodes were removed, and single cell suspensions were prepared. After 48 hours of in vitro culture, interferon-¦Ã (ifn-¦Ã), interleukin-10 (il-10), and il- in the culture supernatant were detected by ELISA. 4. Tumor necrosis factor-¦Á (tnf-¦Á) content; under the induction of type II collagen, normal rat lymph node cells and spleen cells were co-cultured with different concentrations of chinonol to detect collagen-induced lymphocyte proliferation and apoptosis. . Results Compared with the model group, the contents of ifn-¦Ã and tnf-¦Á in the splenocyte culture supernatant of the chinonol group were reduced (p<0.05, p<0.01), and the levels of il-10 and il-4 were increased (p<0.05 , p<0.01); the contents of ifn-¦Ã and tnf-¦Á in lymph node cell culture supernatant decreased (p<0.05, p<0.001), and the levels of il-10 and il-4 increased (p<0.05, p<0.001) ; In in vitro experiments, chinonol inhibited the proliferation of type II collagen-specific lymph node cells and promoted the apoptosis of type II collagen-specific lymph node cells and splenocytes in a dose-dependent manner. Conclusion Chinonol can inhibit the secretion of pro-inflammatory factors by type II collagen-specific lymphocytes, promote the secretion of anti-inflammatory factors by collagen-specific lymphocytes, inhibit the proliferation and promote apoptosis of collagen-specific lymphocytes, and exert immunosuppression in multiple ways. effect.
