Abstract
After analyzing the respective advantages and disadvantages of acid extraction and enzymatic hydrolysis for extracting collagen from cow hide, a new method for extracting collagen from cow hide was studied and finally determined – the acid-enzyme compound method. This method has a simple process and can be used in a relatively short time. Within the preparation time, the collagen extraction rate can reach 54.5%, while ensuring the natural triple helix structure of collagen. At the same time, based on the research results on the properties of collagen, a rapid and accurate identification and quality evaluation method for collagen and gelatin – a rapid identification and evaluation method, including polyacrylamide gel electrophoresis, tannin precipitation experiment, Trypsin sensitivity test and in vitro fibrogenic ability test. The acid-enzyme compound method and rapid identification and evaluation method have practical significance for the large-scale production application of collagen extraction process and the identification and quality evaluation of various products on the market.
