Abstract
Abstract: Objective To explore the feasibility of constructing a tissue-engineered three-dimensional bronchial model in vitro using human bronchial epithelial cell lines and human fetal lung fibroblast cell lines as seed cells and collagen gel composite Matrigel as scaffold material, in order to study bronchial epithelial cells and Fibroblast interactions provide a model. Methods Bronchial epithelial cells and lung fibroblasts were co-cultured in liquid type I collagen added with Matrigel, and a static stretching system was used to construct a bronchial model in vitro. The growth of cells and the three-dimensional tissue structure formed were identified through gross observation, phase contrast microscopy, HE staining and immunohistochemical staining (broad-spectrum CK, etc.). Results: A tissue-engineered three-dimensional bronchial model can be constructed by applying a static stretching system in vitro. Gross observation shows well-shrunk tissue sheets with a certain degree of toughness; the formation of a reticular structure can be observed under a phase contrast microscope, with good cell activity; HE staining shows the formation of a three-dimensional reticular structure; broad spectrum immunohistochemical staining CK and Vimentin were positive. Conclusion A tissue-engineered three-dimensional bronchial model was successfully constructed in vitro. The co-culture of seed cells can complete the polar growth process in the scaffold material and form a network structure.
