Experimental Study On Influence Of Phenytoin Sodium On Wound Healing In Rats

Abstract
Objective To observe the effect of phenytoin on the wound healing process in experimental rats and explore its mechanism of action. Methods 100 SD rats were randomly divided into 5 groups, namely blank control group, lithospermum oil positive control group, phenytoin sodium low-dose group (5 mg/cm~2), and medium-dose group (10 mg/cm~2). , high dose group (20 mg/cm~2), 20 animals in each group. A 2 cm ¡Á 3 cm rectangular wound model was designed on the dorsal side of the rat spine. The wound healing rate was observed on days 4, 8, 12, and 16 after trauma, and immunohistochemistry was used to detect the expression of type I collagen, vascular endothelial growth factor (VEGF), Bcl-2, and Bax in the wound. Results The wound healing rate of the low-dose phenytoin group was significantly better than that of the blank control group and the medium- and high-dose phenytoin groups at 8, 12, and 16 days after trauma (P<0.05). The positive expression of type ¢ñ collagen, VEGF, Bcl-2, and Bax showed a gradually increasing trend. The expression of Bax increased most obviously in the phenytoin sodium medium and high-dose groups, and the difference between the high-dose group and the low-dose group, blank control group, and lithospermum oil group was statistically significant at 16 days after trauma (P<0.05). The expression of type ¢ñ collagen increased the least in the high-dose phenytoin sodium group, and there was a statistically significant difference between the high-dose group, the low-dose group, the blank control group, and the lithospermum oil group 16 days after trauma (P<0.05). On the 4th day after trauma, the expression of VEGF in the phenytoin sodium low-dose group increased significantly. On the 16th day, the expression of VEGF in the lithospermum oil positive control group and the phenytoin sodium low-dose group had no statistical significance, but there were differences compared with other groups. Statistical significance (P<0.05). On the 4th day after trauma, the expression of Bcl-2 in the phenytoin sodium low-dose group and the lithospermum oil group increased simultaneously, and was still higher than that in the other groups on the 16th day (P<0.05). The expression of the phenytoin sodium high- and medium-dose groups was lower than that in the blank control group. (P<0.05). Conclusion The low-dose phenytoin sodium group can promote wound healing by promoting fibroblast proliferation, vascular regeneration, and extracellular matrix deposition.
