Abstract
[Purpose] The Rv3194c gene encodes the PDZ signaling protein of Mycobacterium tuberculosis. This study explored whether it has adhesin properties. [Method] Rv3194c was expressed prokaryotically, and then the Rv3194c protein was incubated with hyaluronic acid, chondroitin sulfate, and type I collagen in buffers at different temperatures (37, 38, 39, 40¡ãC) overnight, and then used Western blot and ELISA to analyze the changes in component content in the supernatant. [Results] Western blot identification results showed that His-Rv3194c protein was expressed in a soluble form, and the molecular mass of the protein was 35 kDa. Western blot showed that the His-Rv3194c protein content in the supernatant of the 39¡ãC experimental group (***P<0.001) was significantly smaller than that of other experimental groups (37, 38, 40¡ãC); ELISA showed that the content of His-Rv3194c protein in the supernatant of the 39¡ãC experimental group The contents of hyaluronic acid (HA), chondroitin sulfate (CS), and type I collagen (Collagen I) (***P<0.001) were extremely significantly lower than those of other experimental groups (37, 38, and 40¡ãC). [Conclusion] It was confirmed for the first time that Rv3194c protein has adhesin properties and can become a target protein for the development of new anti-tuberculosis drugs.
