Abstract
Acid-soluble collagen (ASC) was extracted from the body wall of Urechis unicinctus using 0.5 mol/L acetic acid extraction combined with NaCl precipitation method, and its physical and chemical properties were preliminary studied. Ultraviolet spectrum analysis shows that the characteristic absorption wavelength of ASC is located at 228nm, and it is preliminarily inferred that the extracted protein is a typical collagen; amino acid composition analysis shows that ASC is a typical type ¢ñ collagen; polyacrylamide gel electrophoresis (SDS-PAGE) results show that ASC has two different ¦Á chains (¦Á1 and ¦Á2 chains), ¦Â chain and ¦Ã chain, and its collagen contains disulfide bonds; Fourier transform infrared spectroscopy (FTIR) analysis shows that it has a triple helix structure; differential scanning calorimetry DSC analysis shows that the thermal denaturation temperature (td) and heat shrinkage temperature (ts) are 33.6¡ãC and 67.5¡ãC respectively.
