Abstract
The use of organic acids (acetic acid, citric acid, lactic acid) to extract collagen from the skin of several major freshwater fishes, including silver carp (hypophthalmichthysmolitrix), bighead carp (aristichthysnobdis), and grass carp (glenopharyngodonidellus), was studied. The study found that the protein contents of silver carp, bighead carp and grass carp skins were approximately 25.9%, 23.6% and 29.8% respectively, and the hydroxyproline contents were 1.8%, 1.9% and 2.3% respectively. The results show that when silver carp and bighead carp skins are treated with 2.5% naci solution (1:10), and grass carp skins are treated with 5% nacl solution (1:10) at a temperature of about 5¡ãC and magnetic stirring for 12 hours twice, the soluble impurity proteins can be removed. They accounted for 5.9%, 6.7% and 9.1% of the original total protein respectively. After the fish skin is swollen with acid (1:30) and homogenized, a crude extract of fish skin collagen can be obtained. The acid extraction recovery rates of silver carp and grass carp skins are highest with acetic acid, which are 78.9% and 78.9%, respectively. 84.1%, and citric acid is the highest in bighead carp skin, about 82.0%. The crude extract is then subjected to suction filtration, salting out, dialysis, and finally freeze-drying to obtain collagen products with higher purity. Among the three types of fish skin collagen products, grass carp is the best, while silver carp and bighead carp are slightly worse.
