Abstract
4. Using yak hide as raw material, acid-soluble collagen (ASC) and enzyme-soluble collagen (PSC) were extracted, and the structure and properties of collagen molecules were analyzed. The results show that the extraction rates of ASC and PSC are (54¡À0.18)% and (78¡À0.42)% respectively; through ultraviolet absorption spectrum (UV) analysis, infrared absorption spectrum analysis (FTIR) and electrophoresis analysis (SDS-PAGE), It shows that ASC and PSC have a complete triple helix structure, consistent with the structural characteristics of type I collagen; amino acid analysis found that ASC and PSC are rich in imine, and their contents are 264.1 residues/1000 residues and 276.6 residues/1000 respectively. residue; thermal stability analysis shows that the thermal denaturation temperatures of ASC and PSC are 41.5 ¡æ and 47.5 ¡æ, and the heat shrinkage temperatures are 62.5 ¡æ and 70 ¡æ, proving that the thermal stability of PSC is higher than ASC; ASC was observed with a scanning electron microscope (SEM) and PSC morphology, showing that its surface is a loose, irregular fiber network shape; self-assembly experiments show that both have a certain fiber-forming ability. The D-periods of the self-assembly products are (68.2¡À5) nm and (69.3¡À) respectively. 3) nm, and PSC is faster to assemble than ASC
