Abstract
The collagen content in the dermis of mammalian skin accounts for about 70%, mainly type I, type III and a small amount of type V collagen. The total collagen in rat skin was extracted using dilute acid dissolution and enzymatic methods, and the collagen was The crude extract was denatured at 60¡ãC and then degraded with trypsin. The characteristic peptides of the two collagens were analyzed by liquid chromatography/mass spectrometry, and the relative contents of type I and type III collagen in the skin of rats at different growth stages were studied. Results It shows that the relative content of type III collagen in rat skin gradually decreases with the extension of the growth period, while the relative content of type I collagen gradually increases, and the ratio of the two collagens tends to be stable after 8 weeks. It shows that the use of high-efficiency liquid It is feasible to analyze collagen types and their dynamic changes in tissues using chromatography/mass spectrometry, providing an experimental basis for better clinical applications.
