Abstract
Objective: To explore the effect and mechanism of procyanidin from vaccinium (PC) on myocardial fibrosis in rats. Methods: The rat myocardial fibrosis model was established by subcutaneously injecting isoprenaline (Iso) 5 mg¡¤kg-1¡¤d-1 on the back for 7 days, and PC100, 200, and 400 mg¡¤kg-1¡¤d- were administered by gavage at the same time. 1. Take the drug continuously for 14 days. After the experiment, the heart weight index (HW/BW) and left ventricular mass index (LVW/BW) of the rats are measured; the serum angiotensin ¢ò (angiotensin ¢ò, Ang ¢ò), type ¢ñ and type ¢ó are measured by ELISA. The content of collagen; the change of nitric oxide (NO) content in serum was detected by colorimetric method with microplate reader; the content of malondialdehyde (MDA) in left ventricular myocardial tissue was detected by spectrophotometry; fully automatic biochemical instrument Detect the levels of lactate dehydrogenase (lactatedehydrogenase, LDH) and creatine phosphokinase (CK) in serum. Results: Compared with the control group, the heart mass index and left ventricular mass index of the rats in the Iso model group increased, the levels of Ang II, type I, and type III collagen in the serum increased, the MDA content in the left ventricular myocardial tissue increased, and the NO content in the serum decreased. LDH and CK contents increased (P<0.05 or P<0.01). Both PC200 and 400mg¡¤kg-1¡¤d-1 can reduce the cardiac mass index and left ventricular mass index, and reduce the levels of Ang¢ò, type I and type III collagen in the serum, while PC100, 200 and 400mg¡¤kg-1¡¤d-1 can increase The NO content in serum, reduced MDA content in myocardial tissue, and reduced serum LDH and CK content were statistically different from the model group (P<0.05 or P<0.01). Conclusion: PC can inhibit the synthesis of collagen by reducing the level of Ang II, increasing the level of NO and its antioxidant effect, thereby inhibiting myocardial fibrosis in rats and having a certain protective effect on the myocardium.
