Abstract
Objective To observe the effect of sinomenine (SN) on the DNA binding activity and nuclear translocation of nuclear factor-¦ÊB (NF-¦ÊB) in synovial cells of rats with collagen-induced arthritis (CIA), in order to elucidate this Possible mechanisms of anti-inflammatory drugs. Methods CIA rats were used as models, and synovial cells from knee joints were collected and cultured in vitro. Normal control group, CIA group, CIA+methotrexate 10 ¦Ìmol/L (positive control group), CIA+SN 50 ¦Ìmol/L group, and CIA+SN 500 ¦Ìmol/L were established. Group L. Fluorescent labeling and confocal laser scanning microscopy were used to detect nuclear translocation of NF-¦ÊB p65 subunit in synovial cells, and electrophoretic mobility change assay (EMSA) was used to detect the binding activity of NF-¦ÊB to DNA sequences. Results Compared with the normal control group, obvious nuclear translocation of p65 subunit was seen in CIA rat synoviocytes, and the DNA binding activity of NF-¦ÊB was significantly increased (P<0.01). When SN was at 50 ¦Ìmol/L and 500 ¦Ìmol/L, It can inhibit the nuclear translocation of p65 subunit and the DNA binding activity of NF-¦ÊB in CIA rat synoviocytes in a concentration-dependent manner. Methotrexate can downregulate the DNA binding activity of NF-¦ÊB but does not inhibit p65 at 10 ¦Ìmol/L. Nuclear translocation of subunits. Conclusion SN inhibits NF-¦ÊBp65 nuclear translocation and NF-¦ÊB DNA binding activity in synoviocytes, which is one of its anti-inflammatory mechanisms.
