Interleukin 1#Beta# Inhibits Collagen Synthesis In Cardiac Fibroblasts And Promotes Its Decomposition

Abstract
This article applies ~(3)H–thymidine (~(3)H–thymidine, ~(3)H–TdR) incorporation method and ~(3)H–proline (~( 3)H–proline, ~(3)H–Pro) incorporation method to observe the effect of interleukin–1#beta#, IL–1#beta# on Spague–Dawley milk Effects on DNA and collagen synthesis in mouse cardiac fibroblasts, and using gelatin zymography and Western blot to detect matrix metalloproteinases (MMPs) MMP–2, MMP–9 activities and MMP–2 and MMP–9 For protein expression, RT-PCR was used to detect the mRNA expression of MMP–2 and MMP–9. The results showed that: (1) After different concentrations of IL–1#beta# (0.1, 1, 10, 100 ng/mL) acted on cells for 24 h, the amount of H–TdR incorporated in each group was significantly higher. The control group was lower, and the incorporation amount of ~(3)H–Pro was significantly reduced; while 0.01 ng/mL IL–1#beta# acted on CFs, and the incorporation amount of ~(3)H–TdR was significantly reduced. ~(3) The amount of H–Pro incorporated has no significant effect; (2) Different doses of IL–1#beta# stimulate the activity of MMP–2 and MMP–9 to increase in a dose-dependent manner. IL–1#beta# increases MMP–2 protein expression, respectively (0.01 ng/L: 1.35¡À0.05 times; 0.1 ng/mL: 1.56¡À0.10 times; {sl P}<0.05; 1 ng/m: 2.15¡À0.32 times; 10 ng/mL: 2.34¡À0.19 times; 100 ng/mL: 2.41¡À0.26 times; {sl P}<0.01), while increasing MMP--9 protein expression (0.01 ng/mL: 1.38¡À0.20 times; {sl P}<0.05; 0.1 ng/mL: 2.37¡À0.21 times; 1 ng/mL: 2.56¡À0.31 times; 10 ng/mL: 2.60¡À0.19 times; 100 ng/mL: 2.67¡À0.15 times; { sl P}<0.01); (3) The activities, mRNA and protein expressions of MMP--2 and MMP--9 were all higher than those in the control group. IL--1#beta# stimulates MMP--2 mRNA expression to increase (0.01 ng/mL: 1.81¡À0.16 times; {sl P}<0.5; 0.1 ng/mL: 2.17¡À0.19 times; 1 ng/mL: 2.18 ¡À0.04 times; 10 ng/mL: 2.27¡À0.06 times; 100 ng/mL: 2.31¡À0.07 times; {sl P}<0.01) and increased MMP--9 mRNA expression (0.01 ng/mL: 1.36¡À0.13 times ; {sl P}<0.05; 0.1 ng/mL: 1.49¡À0.08 times; 1 ng/mL: 1.50¡À0.11 times; 10 ng/mL: 1.51¡À0.24 times; 100 ng/mL: 1.52¡À0.31 times; {sl P}<0.01). The above results show that IL--1#beta# reduces the synthesis of collagen by reducing the cell division of cardiac fibroblasts, and at the same time promotes the transcription and post-transcriptional expression of MMP--2 and MMP--9 to promote the decomposition of collagen.
