Abstract
The kinetic properties of collagenase extracted from bacillus cereus mbl13 in enzymatically hydrolyzing bovine bone collagen were studied. The effects of the five factors of substrate mass concentration, time, temperature, enzyme mass concentration, and pH value on the degree of enzymatic hydrolysis were investigated, the conditions for enzymatic hydrolysis were optimized, and the kinetics of the enzymatic hydrolysis process were analyzed. The results showed that the optimal enzymatic hydrolysis process parameters were substrate mass concentration 30g/l, time 6h, temperature 45¡æ, initial enzyme solution mass concentration 0.35g/100ml, ph8.0. On this basis, the enzymatic hydrolysis kinetic equation was derived as: v=9.3633[s]/(114.785+[s]). The dynamic changes of the enzymatic hydrolysis process were measured at wavelengths of 230nm and 570nm, which showed that the collagen peptide content continued to increase.
