Abstract
? This paper studies the purification methods of fish bone collagen from shark, silver carp, grass carp and tilapia and conducts a preliminary study on its characteristics. The sds-page electrophoresis results showed that the purity of the four extracted collagens was high. In comparison, shark bone collagen had the highest purity. Through proteolysis studies, it was found that all four types of fish bone collagen can be hydrolyzed by trypsin and proteinase K, and the hydrolyzate products have different molecular weights and complex compositions. The results of the enzymatic protein spectrum showed that there were certain differences in the electrophoresis patterns of fish bone collagen from different fish species. The closer the fish species are, the smaller the difference in the electrophoresis patterns of the extracted collagen after hydrolysis; conversely, the greater the difference in fish species, the greater the difference in the electrophoresis patterns after hydrolysis of collagen. Amino acid composition analysis shows that glycine is the most abundant in shark and silver carp bone collagen, followed by alanine, glutamic acid and arginine.
