Abstract
Abstract: Objective To observe the preventive and therapeutic effect of dexamethasone on pulmonary fibrosis in rats induced by blemase. Methods 32 rats were randomly divided into control group (n=8), treatment group (n=12) and injury group (n=12). The treatment group and injury group were injected with blemasein into the trachea to prepare lung acute Injury model, the control group was injected with distilled water. After modeling, the treatment group was treated with daily intraperitoneal injection of dexamethasone, while the injury group and control group were given normal saline every day. The rats were sacrificed at different times (3d, 7d, 14d, and 27d) and then underwent alveolar lavage, Wright’s staining for cell classification, HE staining of the lung tissue, and determination of the total amount of collagen in the lung tissue. Results The pathological sections showed that the alveolar intervals of the rats in the injury group were significantly widened, with more fibroblast proliferation and inflammatory cell infiltration, and lung tissue consolidation in the later stage. The pathological changes in the treatment group were significantly lighter than those in the injury group, and the alveoli in the later stage were significantly reduced. The wall has a tendency to recover. The levels of neutrophils in the alveolar lavage fluid of rats in the injury group and the treatment group reached the peak on the 7th day, but the treatment group was lower than the injury group (P<0.05), and it was significantly lower than the injury group on the 14th day (P< 0.01); the total amount of lung collagen in the injury group and the treatment group reached the peak on the 14th day, but the injury group was significantly higher than the control group (P<0.01), and the treatment group was significantly lower than the injury group (P<0.01) . Conclusion Dexamethasone can effectively prevent and treat pulmonary fibrosis by inhibiting the aggregation and infiltration of pulmonary inflammatory cells in lung tissue and the increase in collagen secretion caused by fibroblast proliferation.
