Omentin Can Reduce Collagen Iii Content In Aorta Of Diabetic Arteriosclerosis Rats

Abstract
Objective To explore the effect of omentin overexpression on aortic collagen content in diabetic arteriosclerosis rats. Methods 60 1-month-old male Wistar rats were selected and divided into normal control group (NC group; n=8) and experimental group (n=52) according to the random number table method. The experimental group was fed with high-fat and high-sugar feed combined with tail vein injection of 2% streptozotocin (30 mg/kg) to establish a diabetic rat model; the modeled diabetic rats were then intragastrically administered with vitamin D3 (700,000 IU/kg). kg), continued feeding for 16 weeks to establish a rat model of diabetes combined with arteriosclerosis. The final 24 diabetic arteriosclerosis rats were divided into diabetic arteriosclerosis group (DAC group; n = 8), diabetic arteriosclerosis + empty virus group (DAC + E group; n = 8) and diabetic arteriosclerosis + Omentin group (DAC+O group; n=8). 150 ¦ÌL of adeno-associated virus carrying human omentin gene ITLN1-AAV9 and virus carrying empty plasmid were injected into the DAC+O group and DAC+E group respectively through the tail vein, and an equal amount of normal saline was injected into the NC group and DAC group. Continue to feed high-fat feed for 4 weeks, then sacrifice the rats. Serum and liver omentin and blood lipid levels are measured. The aorta is taken for HE staining. Quantitative real-time polymerase chain reaction is used to determine collagen ¢ñ, ¢ó, matrix metalloproteinase 2 (MMP- 2), MMP-9 mRNA content; Western blot determination of aortic collagen I, III, MMP-2, MMP-9 protein content; xanthine oxidase method to determine aortic superoxide dismutase (SOD) content, thiopar Aortic malondialdehyde (MDA) levels were determined by the bituric acid condensation method. Results (1) Compared with the NC group, the rat omentin levels in the serum of diabetic arteriosclerosis rats were significantly decreased. After intravenous injection of adeno-associated virus carrying the human omentin gene, the human omentin in the serum of rats in the DAC+O group was (101.0¡À0.2) ¦Ìg/L, and the human omentin in the liver was (98.3¡À 1.9) ¦Ìg/L, human omentin was not detected in the corresponding tissues of other groups. (2) Compared with the DAC group, the serum lipid profile and aortic MDA in the DAC+E group and the DAC+O group were significantly decreased, and SOD was increased, but there was no statistically significant difference between the latter two groups. (3) In the DAC group, the aortic intima was partially detached, smooth muscle hyperplasia, local hyaline degeneration and calcification. In the DAC+O group, the intima was intact, the local wall thickened, and the degree of smooth muscle hyperplasia and calcification was significantly improved compared with the DAC group. (4) Compared with the DAC+E group, the collagen ¢ñ and ¢ó mRNA expressions in the DAC+O group were significantly decreased, and the collagen ¢ó protein expression was significantly decreased. Compared with the DAC+E group, there was no statistically significant difference in the mRNA and protein expression of MMP-2 and MMP-9 in the DAC+O group. Conclusion In rats with diabetic arteriosclerosis, overexpression of human omentin can reduce the expression of collagen III in the aorta and alleviate arteriosclerosis without significant effects on oxidative stress levels.
