Abstract
? Pepsin was used to extract collagen from fish scales, and the effects of extraction medium, pretreatment, stirring, extraction times and extraction time on collagen extraction were investigated. The results show that the best extraction process of fish scale collagen is to use microwave treatment to assist in the removal of impurities. After decalcification with 10% citric acid, use 1% citric acid as the extraction medium while stirring, and extract three times, each time for 48 hours. The purified fish scale collagen was analyzed by SDS-PAGE gel electrophoresis and Fourier transform infrared spectroscopy, and compared with the standard type I collagen, it was determined that the extracted protein was a typical type I collagen and was electrophoretically pure.
