Abstract
By investigating the effects of pH, culture temperature, dissolved oxygen concentration and induction timing on cell growth and human-like collagen II expression, the optimal control range of these factors was determined and the fermentation conditions were optimized. The results show that controlling the initial pH to 6.5, the post-induction pH to 6.8, the culture temperature of 34¡ãC, DO 20% and induction in the late logarithmic growth phase are beneficial to cell growth and expression of exogenous genes, and the final cell density is 88.4 g. /L, the human-like collagen II production reaches 14.2 g/L.
