Abstract
Collagen film (containing 87.5 mgi type collagen) was used as a carrier, and 3.5 mgrhbmp-2 (human genetic recombinant bone morphogenetic protein-2) was compounded to prepare a collagen/bmp composite material. The composite material was first embedded in the latissimus dorsi muscle of rabbits to prestructure new bone tissue, and the osteogenesis process and tissue morphology of the composite material were observed using ALP staining, vonkossa staining and HE staining. The formed new bone tissue was then free-transplanted to repair the autologous mandibular body hole defect; direct repair with collagen-based rhbmp-2 composite bone repair material was set as the control group, and the non-repaired bone defect group was set as the blank group. X-rays, compressive strength, hard tissue sections, tetracycline fluorescence staining, and bone morphometry were used to observe the quality and effect of composite materials in repairing bone defects. The results show that the collagen/bmp composite material forms bone in rabbit latissimus dorsi muscle in 4 to 6 weeks, and the collagen material degrades in 3 to 5 weeks; the osteogenesis process is mainly cartilage osteogenesis, and the new bone shape is woven. Bone, obvious microvascular distribution can be seen; free transplantation repaired the autogenous mandibular defect, and the defect area showed bony healing after 6 weeks. There was a significant difference in compressive strength (p=0.041) and new bone volume (p=0.034) compared with the control group. difference. The new bone tissue formed by collagen/bmp composite material in skeletal muscle can be used as bone donor to repair a certain range of bone defects.
