Abstract
Using konjac glucomannan microspheres as the matrix, the microspheres were aminated with 2-diethylaminoethane hydrochloride (DEAE), activated with 1,4-butanediol diglycidyl ether, and the collagen was The protein was coated on the microspheres and then cross-linked with glutaraldehyde to obtain uniformly coated and stable charge-collagen dual-action microcarriers. The collagen coating conditions and activation methods, collagen from different sources, and the amount of DEAE were investigated. On the effect of microcarrier cell culture, the culture of Vero cells on microcarriers was studied. The results showed that the collagen layer of charge-collagen dual-action microcarriers was controllable in the range of 5~50mm, and the adhesion of cells after 2 hours of culture was The amount is about 4¡Á104mL-1, which is between Cytodex1 and Cydodex3. After 96h, the growth rate of cells on the microcarrier gradually exceeds that of the charge-type microcarrier Cydodex1. At 144h, the number of cells is greater than that of Cydodex1 and Cydodex3, reaching 1.59¡Á106mL-1.
