Abstract
In order to enzymatically digest low-value protein resources with bacterial extracellular proteases to prepare antioxidant active peptides and discover new proteases, a liquid fermentation culture method was used to ferment Pseudoalteromonas sp. SHK1-2 to produce enzymes and obtain crude extracellular proteases. The enzyme solution is used to enzymatically hydrolyze dace collagen extracted by hot water method. Through ultrafiltration and sephadex LH-20 molecular sieve chromatography, it is obtained with DPPH free radical scavenging ability (35.6% ¡À 7%) and oxygen free radical scavenging ability (Oxygen radical). absorbance capacity (ORAC) and DNA oxidative damage inhibitory activity. Liquid chromatography-mass spectrometry identified the molecular weight of this active peptide as 776.2 Da, and the predicted amino acid sequence is Thr-Ala-Gly-His-Pro-Gly-Thr-His . ORAC testing has verified that synthetic peptides also have antioxidant activity. Research shows that bacterial extracellular proteases have certain application prospects in increasing the value of low-value resources, and it has certain reference significance for the exploration of new proteases and their applications.
