Preparation Of Free Radical Scavenging Peptides From Nile Tilapia Skin Collagen And Its Antioxidant Activity

Abstract
Bromelain and alcalase enzymes were used to perform composite enzymatic hydrolysis of Nile tilapia skin collagen. The in vitro antioxidant activity of the enzymatic hydrolyzate was studied. Experiments show that the enzymatic hydrolysis product has strong superoxide anion/hydroxyl radical scavenging activity and reducing ability. Ultrafiltration membranes with different molecular weight cutoffs are used to separate the enzymatic hydrolyzate into five components, namely tgh-¢¡ (>10ku), tgh-¢¢ (10¡«5ku), tgh-¢£ (5¡«3ku), tgh-¢¤ (3¡«1ku) and tgh-¢¥ (10ku). Among them, the tgh-¢¥ component showed the strongest superoxide anion radical scavenging activity, so gel filtration, ion exchange and reverse high-pressure liquid chromatography were used to further separate and purify this component. The purified peptide has strong superoxide anion radical scavenging activity, and its ic50 value is 4.6 ¦Ìg¡¤ml-1. Mass spectrometry analysis showed that the molecular weight of the peptide ranged from 311.3 to 932.8u.
