Abstract
Using tilapia skin as raw material, the enzyme method was used to prepare collagen peptide products. The basic wood composition of tilapia skin was determined, and the low-temperature continuous phase change extraction technology was used for degreasing. The degree of hydrolysis, molecular weight of hydrolyzate and sensory evaluation were used as indicators to study the influence of protease types on enzymatic hydrolysis. The degree of hydrolysis was used as an indicator. , study the effect of enzymatic hydrolysis conditions on the degree of hydrolysis, and use response surface experiments to optimize the preparation process of fish skin collagen peptide; use gel filtration chromatography to determine the molecular mass distribution of the enzymatic hydrolysis solution. The results show that the use of alkaline protease to hydrolyze fish skin has the best effect. The optimal process conditions for enzymatic hydrolysis are: time 7 h, enzyme addition amount 7%, temperature 60 ¡æ, pH value 10.38. Under these conditions, alkaline protease hydrolyzes rostrogen. The hydrolysis degree of non-fish skin can reach up to 23.01%. Collagen peptides obtained by this method are mainly oligopeptides (about 2 to 8 amino acid residues) with a molecular mass between 180 and 1000 u. They have good physiological activity and can be widely used in health foods as functional factors. , cosmetics, medicine and other industries. The collagen peptides from tilapia skin has been extracted by using an enzymatic method. The composition of tilapia skin was also analyzed. A novel continuous phase-transition extraction method at low temperature was used to remove fat from tilapia skin. The degree of hydrolysis, molecular weight and organoleptic investigation were used as indicators to study the effects of protease types on the hydrolysis. Response surface experiment was designed to optimize the preparation technology of the collagen peptide. GFC was used to determine the molecular weight distribution of hydrolysates. The results showed that alcalase was the best enzyme for the hydrolysis, and the optimal hydrolysis conditions were determined as follows: enzymolysis time 7 h, alcalase dosage 7%, temperature 60 ¡æ, and the pH 10.38. Under these conditions, the maximum hydrolysis degree was 23.01%. The peptides obtained is mainly composed of peptide with 2 to 8 amino acid residues, and has good physiological activity. It could be widely used as a functional factor in various industries, such as health food, cosmetics, medicine and other industries.
