Abstract
In order to improve the production of recombinant human type III collagen fermented by recombinant Pichia pastoris, the composition of BMGY medium (Buffered minimal glycerol-complex medium) during its growth stage was optimized using a response surface. Through the Placket-Burman test, it was concluded that yeast extract, peptone, and glycerol have a significant impact on collagen production. Through the Box-Behnken center combination experiment and Design-Expert analysis software, a response equation with collagen production as the response value was established, and we obtained The optimal concentrations were 1.13% yeast extract, 1.61% peptone, and 0.86% glycerol. Experimental verification showed that after culturing for 12 hours under optimal BMGY medium conditions, the dry weight of Pichia pastoris was 4.41 g/L, and the weight of the bacteria in the growth stage increased by 26%. Through high-density fermentation in a 22 L fermentation tank, the yield reached 4.71 g/L. This recombinant collagen has a significant repair effect on the gastric mucosa burned by acetic acid in rats, laying the foundation for the application and preparation of biomedical materials.
