Abstract
In the collagen sol-gel system with a simulated pH value of 7.4 in vivo, ¦Â-sodium glycerophosphate (¦Â-GP) was added as the phosphorus source for mineralization, and polyacrylic acid (PAA) and sodium tripolyphosphate (TPP) were introduced as The “soft template” simulates the N-terminal and C-terminal isolated functional domains of the non-collagen protein DMP-1 in vivo, regulates the mineralization of type I collagen, and constructs a nano-hydroxyapatite/collagen (n-HA/COL) composite scaffold material. Human umbilical cord mesenchymal stem cells hUMSCs were used as seed cells to explore their effects on cell adhesion, proliferation, and differentiation. The results show that collagen fiber organization and mineralization proceed simultaneously, and aging treatment is beneficial to the crystallization of hydroxyapatite (HAp) crystals. Self-assembled HAp nanoparticles are not only deposited on the outside of collagen fibers, but can also be found on procollagen molecules and at the ends of collagen microfibers. At the same time, nanocalcium phosphate salts are distributed along the cylindrical surface of collagen microfibers. Cell culture showed that n-HA/COL did not significantly promote the proliferation of hUMSCs cells, but could induce the differentiation of hUMSCs into osteoblasts and promote the expression of AKP in osteoblasts. Moreover, n-HA/COL composite scaffold material and osteogenic induction solution ( The induction effect is more significant when combined with OICM). This “bottom-up” deep mineralization method provides a new idea for the preparation of bone scaffold materials. The n-HAp/collagen composite material is expected to be used for bone tissue filling and repair.
