Research On New Extraction Technology Of Catfish Skin Collagen

Abstract
The effect of swelling catfish skin with acetic acid solution on the extraction rate of collagen from the fish skin was studied. It was found that the fish skin was first swollen with 0.5mol/L acetic acid for 6 hours, then beaten with a tissue masher, dissolved with dilute acetic acid, and finally dissolved in alkali solution. And precipitation to obtain crude collagen, the collagen extraction rate can reach 92%, the extraction process is simple, the extraction rate is high, and the time is short. Five proteases were used to hydrolyze the extracted crude collagen to remove impurity proteins. Among them, trypsin was more effective, with an impurity removal rate of more than 60%. The desalination effect of crude collagen by water washing method was studied. SDS-PAGE of the extracted collagen showed that its ¦Á chain was not degraded during the extraction process. The circular dichroism spectrum and X-ray diffraction results of the collagen showed that the extracted collagen basically remained intact. Triple helix structure without denaturation
