Research On The Application Of Element Balance And Metabolic Balance In The Expression Phase Of Human-Like Collagen

Abstract
The elemental balance and metabolic balance methods were applied to establish a mathematical model for the expression phase of human-like collagen produced by fermentation using recombinant Escherichia coli, and the nonlinear optimization method was used to estimate the unknown parameters in the model. The results show that the model is consistent with the growth and metabolic kinetics model of recombinant E. coli, and the key calculation parameters ¦Áh, ¦Áp and mx of the model are 1.173 molo C-mol-1, 293.814 molo C-mol-1 and 17.878 molo C- respectively. mol-1oh-1. This model can better predict the macroscopic reaction rate during the expression period of recombinant human-like collagen, and during the fermentation and expression period of human-like collagen, the growth of the bacteria must be controlled by controlling the glucose feeding rate.
