Abstract
This article aims to use response surface methodology to optimize the extraction process of collagen from horse-faced fish skin and analyze its amino acid composition, so as to provide a certain theoretical basis for the deep processing and high-value utilization of horse-faced fish skin. The results show that the pre-treatment conditions for collagen extraction are: 10% isopropyl alcohol is used for degreasing, the material-to-liquid ratio is 1:15, and the time is 20 hours; 7.5% NaCl solution is used to remove impurity proteins, the material-to-liquid ratio is 1:10, and the time is 24 hours. The optimal extraction process of collagen is: 0.38mol¡¤L-1 acetic acid solution, solid-liquid ratio 1:30, time 33h. Under these conditions, the actual yield is 362.5mg¡¤g-1. Amino acid analysis showed that the glycine content was 24.12%, and its residues accounted for about 1/3 of the total residues. The hydroxyproline content was 9.31%, and tryptophan was not detected, which is consistent with the characteristics of collagen.
