Response Surface Optimization Of The Extraction Process Of Enzymatic Collagen From Grass Carp Swim Bladder

Abstract
In order to improve the extraction rate of collagen from grass carp scraps and swim bladders, the response surface method was used to optimize the extraction of collagen from grass carp swim bladders by pepsin. The effects of enzyme addition amount, extraction time and liquid-to-material ratio on the extraction rate were discussed; a response surface optimization test was conducted based on the single factor test. The results show that the optimal process conditions for pepsin extraction of grass carp swim bladder collagen are 40 mg/g of enzyme, 48 hours of extraction time, and 80:1 (mL/g) liquid-to-material ratio. Under these process conditions, the actual extraction rate of collagen is 17.82%, which is close to the model predicted value (18.04%). Analysis of variance showed that the model was reliable and all three factors had a significant impact on the extraction rate of collagen from grass carp swim bladder.
