Abstract
Objective To detect the biocompatibility of type ¢ñ collagen (Col-¢ñ)/polyvinylalcohol (PVA) copolymer spinning. Methods: Mouse fibroblasts (L-929) were used and divided into 100% concentration Col-¢ñ/PVA copolymer spinning extract, 50% concentration Col-¢ñ/PVA copolymer spinning extract, blank control, and positive control. 4 groups, MTT method was used to measure the optical density value of cells in each group on 1, 2, 3, 4, 5, 6, and 7 days, calculate the relative growth rate (RGR) of cells, and determine the level of cytotoxicity; direct contact co-culture L-929 cells, inverted microscope and scanning electron microscope were used to observe the growth of cells on Col-¢ñ/PVA copolymer spinning; Col-¢ñ/PVA copolymer spinning and polyethylene terephthalate (PET) fibers were respectively Implanted under the skin of Kunming mice, specimens were taken at 1, 3, and 9 weeks after surgery to grossly observe the capsule condition, and histological observation was performed using HE and Mallory trichrome methods. Results MTT method: The relative proliferation rates of cells in the two experimental groups at different time points were 90% to 123.61%, and the cytotoxicity rating of Col-¢ñ/PVA copolymer spinning was 0 to ¢ñ level; direct contact method: cells and Col -¢ñ/PVA copolymer spinning direct contact co-culture, growth is good, normal shape, and attached growth. Col-¢ñ/PVA copolymer spinning and PET fibers implanted subcutaneously had capsule formation at 1, 3, and 9 weeks, and the transparency of the capsule increased with time. The inflammatory cells around the capsule were mainly neutrophils, The number of macrophages, lymphocytes, and inflammatory cells decreased significantly over time. Conclusion Col-¢ñ/PVA copolymer spinning has good biocompatibility, and its cytotoxicity and histocompatibility are within the allowable range for clinical application.
