Abstract
This paper studies the preparation, isolation and purification of collagen peptides from a low-value sea cucumber of the family Colocaceae – Paracaudina chinens var. and its free radical scavenging activity. Bromelain was used to enzymatically hydrolyze seaweed collagen. Through orthogonal experiments, it was determined that the optimal enzymatic hydrolysis conditions for bromelain to hydrolyze seaweed collagen were: enzymatic hydrolysis temperature 40¡ãC, enzyme addition amount 240u/ml, substrate concentration 18mg/ 100ml (calculated as hydroxyproline), ph5.2, enzymatic hydrolysis time is 4h. Under this test condition, the superoxide anion radical scavenging rate of the product obtained can reach 52.20%. The enzymatic hydrolyzate was separated and purified using ultrafiltration and sephadexg-25 gel column to obtain components with strong ability to scavenge superoxide anions and hydroxyl radicals.
